大量研究表明动物机体对甘氨酸螯合铁有更好的生物利用率,而甘氨酸螯合铁较好的生物学效价可能与其在动物机体内的吸收机制有关。动物机体吸收外源铁主要发生在十二指肠和空肠上段,并需要相关膜转运蛋白的协助。二价金属离子转运蛋白 (Divalent Metal Transporter 1DMT1) 可将肠道内包括Fe2+ 在内的二价金属离子转运到细胞内,其主要分布在动物十二指肠绒毛刷状缘及隐窝等处,其表达变化主要由感知体内铁储存的反馈机制来调控基底膜铁转运蛋白 (Ferroportin 1Fpn1) 主要负责将细胞内的铁转运到血液中。植酸和纤维素等抗营养因子能够与肠道中的亚铁离子反应,形成不溶性复合物,从而影响铁离子在肠道内的吸收;氨基酸螯合铁中的铁元素因为和氨基酸配体的螯合作用,可以避免植酸和纤维素等因素的影响,从而获得更好地吸收效果;在 Caco-2 细胞模型上,抗坏血酸铁和氨基酸螯合铁具有相似的铁吸收机制,均是以铁离子的形式通过细胞中的 DMT1 吸收的。

Effects of Iron amino acid chelate on animal absorption

Many studies have shown that animals have better bioavailability of iron glycine chelate, and the better biological titer of iron glycine chelate may be related to the absorption mechanism in animals. The absorption of exogenous iron mainly occurs in the duodenum and upper jejunum, and requires the assistance of related membrane transporters. Divalent Metal Transporter 1 (DMT1) can transport intestinal divalent metal ions, including Fe2+, into cells, mainly at brush margins and crypts of duodenal villi. The change of its expression is mainly regulated by the feedback mechanism of sensing iron storage in vivo. Ferroportin 1 (Fpn1) is responsible for transporting iron from cells into the blood. Phytic acid, cellulose and other anti-nutrient factors can react with ferrous ions in the intestine to form insoluble complexes, thus affecting the absorption of iron ions in the intestine. The iron element in amino acid chelate iron can avoid the influence of phytic acid and cellulose because of chelating with amino acid ligand, so as to obtain better absorption effect. In the Caco-2 cell model, iron ascorbate and iron amino acid chelate have similar iron absorption mechanisms, both of which are absorbed by DMT1 in the cell in the form of iron ion.
